Welcome to the forum

Try starting in Neutral
Key on, engine off, foot on the brake, shift to N
Then try to start

There is a Neutral switch on the driver's side of transmission, called DTR in 2001, it must be fully in Park or in Neutral to pass starter signal to starter motor
Since Park is at one end of the shifters range, if the cable is out of adjustment then when you shift it into Park on the column it may not move the Neutral switch all the way in to Park, so no start

You tighten the cable mechanism, which is good, but that can also through off the Neutral switch on the other end of the shifter cable, just enough to prevent it from passing the signal to starter motor

Other thing it can be is PATS(passive anti-theft system)
But you didn't mention a rapid Flashing THEFT light on the dash with key on, icon can look like a padlock

I already tried that. I even got under it and made sure shifter was In place with my hand. I unhooked battery cables to make sure it didn't need resetting. Then hooked up my obd scanner and it didn't show anything.

My obd reader did mention the pats but it won't clear it. The lights,radio,a/c and all lights on dash work but won't do anything when try to start it. The 2 bolts I tightened up are under the dash above the brake pedal.

Loosen them up and see if it will start.

check fuse 24 in the Cab fuse box, 7.5amp

So OBD2 reader doesn't turn on?
Or turns on but no connection to computer?

Which PATS code?
B1213 Less than 2 (or 3 if equipped w/Valet key) keys programmed to the system.
B1232/B2103 Transceiver internal antenna damaged. Replace transceiver.
B1342 ECU is defective (EEPROM in PCM is not working — replace PCM). PATS related when stored in PCM only.
B1600 Non-PATS Key or damaged key.
B1601 Unprogrammed encoded ignition key detected (leave ignition on for 20 seconds before trying a programmed key, in Anti-Scan Mode).
B1602 Partial detection of encoded ignition key.
B1681 Transceiver signal not detected.
B2141 Non-Volatile Memory (NVM) configuration failure — No PCM ID stored in PATS.
B2139/U2510 PCM ID does not match between the PCM and PATS control(PATS/ICM/VIC/HEC/SCIL).
B2431 Key program failure (defective key or transceiver).

Yes obd did turn on. Only code it showed was the P1000 I mentioned in other post. The truck won't turn on. I did get the anti-theft reset. When I turned the key forward the back up lights are coming on. So some how it's not all the way in park. Should I have put it in neutral before I tightened up the bolts under the dash?

The transmission is in Park or vehicle would roll away, but the DTR switch is not IN the transmission its ON the outside of transmission, it is moved by the shift cable, which is what you tighten up

There is no reason that tightening those 2 screws/bolts should cause a no crank, but apparently it did from your description, so "we" are just guessing at what may have happened
And I assume you tightened them because column shifter was loose?
"I already tried that" .............that means you shifted to Neutral and still a no crank?
You cab also try starting in R or D but be careful in case it does crank
Checked fuse 24 in cab fuse box?
There is a starter relay in the engine bay fuse box, you can pull it out and then put it back in, that will clean its contacts

There is a THEFT light on the dash, it should come on with Key on and then go OFF, that means key in ignition "passed the test", if it flashes then no crank because of PATS

P1000 means battery was unhooked, it will reset itself after driving a few days, when you can start engine, lol
