Rough Idle, Misfire, Check Engine Light flashed and went away

Hey all, on my 2008 3.0L V6 Sport, I had been driving for maybe 40 minutes. I parked, engine on, and it began to vibrate violently. The CEL flashed. When I put it into drive and drove home (5 minutes) it went away.

Borrowed my buddies OBD scanner, and got this:

It's a cylinder 6 misfire, plus a fuelsystem1 OL Fault . . .

The light is gone, but I want to address this. I have new spark plugs and wires ready to install when I've got time, probably Tuesday. Likewise, I cleaned the MAF just three days ago, did an oil change, and had topped up coolant.

What can I test for? Short of pulling the plug from 6; could it be a coil pack issue? A fuel filter/delivery issue?

Any help would be massively appreciated. Cheers!

OL Fault is likely because the 02 sensor(s) and computer noticed the misfire on 6.

Whatever happened was specific to 6, so is isolated to 6.

Some things to check:

-You could have a bad plug with an enlarged or damaged gap. You can check to see if it's wet, and if so, that means no spark.
-You could have a damaged wire.
-Coil pack runs in pairs, so only three real coils, so if it were the coil, it should affect bank 5 and 6, but if say the coil post for 6 developed a crack, the spark may jump to ground and not reach the wire. (Had this happen personally, I could see the spark jumping down to the valve cover in the dark) You can also reverse the wires for 5 and 6 at the coil and if the misfire moves to 5, the coil post is bad.
-You could have a dead/dying fuel injector on 6
-You could have recessed valve seat issues (Normally an 04-06 3.0 issue and worst case scenario) but a leak down test can verify this. Not likely since this problem seems to be intermittent.
