On the Fuel Rail, where the fuel injectors are, you will see a schrader valve(looks like a tires air valve), should have a cap on it just like a tire's valve would.
Put a towel over it, your hand, when you press down on the center pin, just like letting the air out of a tire, there can be up to 65psi of pressure, so gas can squirt 30 feet.
Dirty Fuel filter usually wouldn't cause a no start, it would cause limited power at highway speeds at first, because of reduced flow, but engine would start and idle fine.
The fuel pump in the tank should come on for 2 seconds each time the key is turned on, this is to prime the fuel pressure.
If possible listen for this to happen, repeat as many times as you want, you can't "over-pressurize" this system.
And if you want to check if it is even a fuel related no start, put some gas or starting fluid in the intake and crank the starter motor.
If it fires up then dies, fuel is your problem
If it doesn't fire up then spark is the problem.
Or on a 2.5l it could be timing belt broke, engine would usually have a lope to it when cranking starter motor if that was the case, i.e. it would sound funny.

We put some starter fluid in the throttle body (removed air tubing and held gas pedal to get it inside body). On first crank after this it tried to start and almost seemed like it did fire and then shut off immediately. Any ideas?

You have spark and timing belt so reads like a dead fuel pump, not filter, but for $10 filters are always good to change
Can you hear pump come on with the key?
If not then there are a couple of things to check before you have to replace the pump.
In the engine compartment fuse box there is a Fuel Pump Fuse, usually 30amp
And there is a Fuel Pump relay, a black cube, put your finger on it when someone turns on the key, you should feel it click inside then click again after 2 seconds so on then off.
Then there is the inertia switch, it cuts power to the fuel pump in case of an accident.
It is located in the passenger side of the cab near passengers feet.
It can be just under the glove box behind the top of the carpet or behind the kick panel(panel just in front of the door on the side), usually at the top.
It will have a reset button on the top.
Truck has to be bounced pretty good to turn off fuel pump this way, or towed, so longshot but not a no shot
