2000 4.0 ohv #6 misfire-have done just about everything

Welcome to the forum
First thing to do when you have a misfire is a Compression test
Remove all spark plugs first
Then test each cylinder and write down the results
If a cylinder 15psi less, or more, than the others then you most likely have a bad exhaust valve
2000 4.0l should have 150psi average but depends on the gauge and battery condition(cranking speed)
The exact numbers are not the point, having a cylinder lower that the average after the test is the point
Changing the spark plugs first is fine, always need new spark plugs, but if problem is still there then don't waste more time and $$$ on other things, test compression to either eliminate it, or confirm it is a compression issue, your wallet will thank you

How long has it been misfiring?
Odometer and transmission type often helps....same for modifications.
Check for evidence of arcing along #6's 'electronic pipeline'.
